The Keeper was a female Wraith responsible for guarding her sleeping brethren on board a hive ship during their centuries-long hibernation. Using her powerful telepathic abilities, she interrogated Colonel Marshall Sumner and from him learned of a rich new potential feeding ground: a planet called Earth. She was killed by Major John Sheppard, which prompted the premature awakening of the entire Wraith population, who now have knowledge of Earth and know that the only way to get to it is through Atlantis. Upon her death, the Wraith Keeper was revealed to also be the Queen of the hive ship... and the Wraith don't take too well to their boss being whacked.
