In the mouth food is chewed; crushed and ground down by the teeth and mixed with saliva until it becomes a moist ball (bolus) of food. In the stomach, food is churned and mixed to a paste called chyme to allow further breakdown by the digestive juices which contain hydrochloric acid (HCl). A vast array of physiological processes has been targeted in an attempt to counter obesity. In France, two medicaments have been approved and are widely used for the treatment of obesity, Sibutral (sibutramine, generic name) and Xenical (orlistat, generic name). First of all there is the brand name or trademark®, generally short, catchy, in the hope that people remember. The generic name sometimes reflects the chemical composition, the origin of the medicament, its function or sometimes nothing. The systemic name is the name agreed upon by the IUPAC (international union of pure and applied chemistry nomenclature).
It also comes under different trade names and one generic name, orlistat, and is produced by two companies.
The above described limited long-term success of behavioural changes (life-style) and of pharmacological intervention in the treatment of severe or morbid obesity has led to a renewed interest in bariatric surgery. To be successful, all surgical procedures described below require strict compliance to diet.
The principle of this procedure is the same as for the gastric band, but this time both a fixed band and staples are used to create the small pouch (figure 6b). Here the reduction of the size of the stomach is accompanied by a bypass of the digestive tract, aiming at a decrease in the absorption of food.
Digestion starts in the mouth where food is degraded by mechanical mastication (chewing) and by alpha-amylase, an enzyme in the saliva (produced by sublingual, submandibular and parotid glands) that breaks down starch (composed of amylose and amylopectin) and glycogen, yielding maltose and short glucose chains. In the stomach the masticated food is mixed with: a) high concentrations of hydrochloric acid (HCl), which main role is to disable potential harmful micro-organisms, b) pepsin and gelatinase, enzymes that break down protein, and c) amylase and lipase. The digestive process gets a real boost in the duodenum, the first 25 cm of the small intestine. Further down, in the small intestine, a last series of enzymes is added, predominantly those that break down sugars in their disaccharide form.
The small intestine is followed by the large intestine (~135 cm), comprising appendix, cecum, colon and rectum. The waste products, a mixture of undigested food, bacterial flora and shed epithelial cells, pass on to the rectum and are expelled from our body through the anus. The digestive process is greatly hampered when a bypass is created (surgically, see figure 8) that leaves out the duodenum and the larger part of the jejunum. Hydrolysis of carbohydrate polymers such as cellulose, xylan and starch is used to produce fermentable sugars for bio-ethanol production. Digestion of biomass material and its fermentation represents the best path forward for bio-ethanol production. The use of lignocellulosic material to produce ethanol presents a number of challenges relative to the use of starch containing sources such as corn, which can be digested with amylases. In order to improve the ethanol yield from these polymers, scientists have turned to the use of specific enzymes from a number of different sources. Xylanase activity was measured using an EnzCheck Ultra Xylanase assay kit as described previously [2]. Cellulase activity was also measured directly using an EnzCheck Cellulase substrate as described previously [2].
In order to demonstrate the utility of microplates and microplate readers, three different commonly used enzymes responsible for the digestion of polysaccharides used as feed stocks for ethanol production were investigated for activity under a number of environmental conditions.
The effect of pH activity was tested on amylase enzyme isolated from three different sources. Xylanase enzyme isolated from the fungi Thermomyces lanuginosus was found to be very temperature stable.

The walls of the alimentary canal have the same four basic layers, (also known as tunics) — the mucosa, submucosa, musclaris externa, and serosa. In certain regions of the alimentary canal, the mucosa may perform one or all three of these functions.
External to the lamina propria is the musularis mucosae, a layer of smooth muscle cells that produces local movements of mucosa. The submucosa, just external to the mucosa, is areolar connective tissue containing a rich supply of blood and lymphatic vessels, lymphoid follicles, and nerve fibers which supply the surrounding tissues of the GI tract wall.
The serosa is the outermost layer of the intraperitoneal organs (it’s also considered the visceral peritoneum). The procedure comprises a small incision (less than 2 cm), near the belly button, the injection of carbon dioxide, to separate the organs of the abdomen, and the insertion of a camera. These are lactase (to split lactose in milk into glucose and galactose), (iso)maltase (to split maltose in two glucoses) and sucrase (to split sucrose in glucose and fructose).
Throughout the passage, different products are added (produced by the body or by bacteria) in order to facilitate the digestive process. Food does not traverse the duodenum or the upper part of the jejunum and this renders the digestive process much less efficient. The ileum (second part of the small intestine) is directly stitched to the proximal part of the duodenum (before the entry of the ducts of the pancreas and the gallbladder). The exogenous digestion of these polymers on an industrial scale can be cost prohibitive without optimization of the enzyme reaction conditions. While starch based feedstocks have been used extensively, it is believed that lignocellulosic energy sources offer a better long-term prospect as a source for fermentable sugars. Using a combination of in vitro digestion, in conjunction with in vivo genetic manipulation of yeast and bacteria strains, the digestion efficiency to fermentable sugars cannot only be increased, but the ability to ferment monosaccharides other than glucose can be improved.
The ability to measure multiple samples rapidly in small volumes offers significant advantages in terms of time, expense and effort.
Carboxymethyl cellulose (cat # 419273), xylanase from Thermomyces lanuginosus (cat# X2753), from Trichoderma longibrachiatum (cat # X2629), cellulase from Aspergillus niger (cat# C1184), α-amylase from Aspergillus oryzae (cat# 10065) from Bacillus licheniformis (cat# A4551) and Bacillus subtilis (Cat#10069) were purchased from Sigma-Aldrich.
In these experiments, equivalent concentrations of a reference standard were used to normalize data prior to plotting.
The direct assay has similar detection limits as the glucose determination, but does not require an overnight digestion. Cellulosic materials do not compete directly with food stocks for resources such as tillable land, fertilizers and water. We also discussed the two main divisions of the digestive system, organs within the alimentary canal and accessory digestive organs. Each layer contains a certain tissue type that plays a crucial role in the breakdown of food. It’s a moist epithilial membrane that lines the alimentary canal lumen from mouth to the anus. Digestive mucosa is made up of three sublayers: (1) a lining epithelium, (2) a lamina propria, and (3) a musclularis mucosae. In the small intestine, this muscle layer’s tone throws the mucosa into a series of small folds that immensely increases its surface area.
Its elastic fibers enable the stomach to regain its normal shape after temporarily storing a large meal.
In most alimentary canal organs, its made up of areolar connective tissue covered with mesothelium, a single layer of squamous epithelial cells. Retroperitoneal organs have both a serosa (facing the peritoneal cavity) and an adventia (on the side abutting the dorsal body wall).
This is possible because the small intestine is very long (around 6 metres) and it is lined with tiny finger like projections called villi. One of the best examples is aspirin; originally the trademark of Bayer AG*, now a generic name for acetylsalicylic acid, a product that goes under many brand names such as Ascriptin, Bufferin, Cama, Easprin, Ecotrin or Empirin.
It is a lipase inhibitor, preventing triglycerides (fat) in our gut to be converted into fatty acids (see figure 3).
In order to make the triacylglycerols more accessible to the lipase they are mixed with bile salts that are produced by the liver and released from the gall bladder. Because of this, one loses the sense of hunger and instead obtains a sentiment of content because of a full stomach (satiety).
Pancreatic products are: trypsin, chymotrypsin, carboxypeptidases and elastase, all of which break down proteins into amino acid fragments, amylase, which breaks down starch and glycogen (and sugar chains that are linked to proteins (glycoproteins)) and nucleases, which break down nucleotides like DNA and RNA. We (the host) provide the food and they (the bacteria) provide the enzymes (released in the gut) that ferment fibrous carbohydrates (cellulose, amylon, amylopectin) into short-chain fatty acids. Pancreatic enzymes and bile salts only enter the jejunum after roughly 100 cm (near the ileum). The duodenum-jejunum is next blocked on one end and connected to the downstream end of the ileum (near the connection with the large intestine). A second cut is made at the border of the jejunum and the ileum (remember, the first and second part of the small intestine respectively). We have previously shown the ability to quantitate these enzymes; here we describe the optimization of some of the reaction conditions for the enzymes that catalyze the digestive hydrolysis of polysaccharide polymers into monomeric constituents.
Lignocellulosic feedstocks, such as wood chips, corn stover or switchgrasses do not directly compete with food sources for land or consumption. The chain structure forms a stiff extended conformation that results in extensive hydrogen bonding with adjacent chains. Termites and ruminant herbivores digest cellulose through the action of symbiotic bacteria located in their intestines and ruminating chambers respectively.
The use of microplates is one way that large numbers of samples can be handled with minimal amounts of reagents and time.
Amylase enzyme isolated from different organisms were compared for their activity at different pH levels. Enzyme concentration titration curves indicate that higher temperatures result in slightly more hydrolysis than ambient temperatures (Figure 7). Using the direct substrate assay, the effect of pH on Aspergillus niger cellulase enzyme activity was investigated.
Grain based feed stock rich in starch requires amylase enzymatic activity, while plant and wood material needs cellulase and xylanase activity to break down the cellulose and xylan material contained in the plant fibers. It can be chemically digested with a combination of high temperature and concentrated acids [3].


Ethanol is currently used as an additive to gasoline in the US to replace MBTE as a means to increase gasoline oxygen content. In this article, we’ll discuss the structural characteristics of the alimentary canal, which is also known as the (GI) gastrointestinal tract. It typically has an inner circular layer and an outer longitudal layer of smooth muscle cells. While the exact cause of IBS is unknown, treatment usually involves medication for gas and diarrhea, as well as changes to diet. I use an adult sized water weighted hoop sometimes if I need more massage, or else just a weighted adult hula hoop. The caecum is a short pouch linking the small intestine to the colon, which is around 1.5m (5 ft) long.
Other herbs will tone and firm the lining of the intestine, preventing the looseness we associate with diarrhoea and leaky gut. Unfortunately, such diet restriction often has substantial negative psychological consequences (a lack of reward signals) and compliancy is generally low (see figure 1). In conclusion, for a large majority, meaningful weight reduction requires a little help, either from a medicament (a really effective one is not yet on the market), an adorable (and expensive) personal coach (whose intimate presence makes up for the lack of food rewards) or a bariatric surgeon.

You happen to travel through a drought- and war-ridden country and come across someone who shows clear signs of starvation. After World War I, as part of the Treaty of Versailles in 1919, Bayer was forced to give up the Aspirin trademark (in France, UK and US), as well as that of Heroin® (another generic name). The active compound, administered orally, is sibutramine, a phenylethylamine derivative (resembling amphetamine) that acts by inhibiting the serotonin-noradrenaline re-uptake transporters (see figure 2). The digestive tract is unable to absorb triglycerides (see below) and they therefore remain in the faeces.
In England, weight-loss surgery has increased 10-fold since 2000, with nearly 7000 operations in 2008. Restrictive surgery aims to accelerate the distention of the stomach wall, thus advancing the onset of satiety. The typical empty stomach has a volume of 100 ml, which, in an adult, can be extended to at least to 1000 ml (figures vary from source to source). Importantly, the small intestine is the major site for absorption of food, ions and water, a process that requires specific transporter proteins. Regardless of the source of feed stock, current production costs make them unpalatable as a fuel. Primary and secondary walls contain cellulose, hemicellulose and pectin, albeit in different proportions.
The result is the formation of microfibrils with high tensile strength and poor water solubility.
Xylose is a five carbon monosaccharide that can form either furanose (5-member) or pyranose (6-member) ring structures. Here we describe the use of microplates to assess physical assay characteristics, such as pH, temperature and enzyme concentration, of some of the enzymes that catalyze the hydrolysis of plant polysaccharides. Correction for the influence of pH on fluorescent signal data was made by normalization each well of the initial unreacted read to a common value. Interestingly, temperatures very near ambient (25° C) had the greatest reduction in enzyme activity (Figure 4) when assessed using the fluorescent amylase substrate. The fluorescence was plotted after 30 minute incubation from assays run at different temperatures.
As seen in Figure 10, Aspergillus niger cellulase exhibits significant activity in acidic pH levels.
Regardless of the enzymatic requirements it is important that the conditions for maximal enzymatic activity be optimized.
However, this technology is currently unfeasible from both an economic and hazardous waste standpoint. In several places along the tract, the circular layer thickens and forms sphincters that act as valves that control food passage from one organ to the next, they also prevent backflow. They can also cause much more severe problems, requiring hospitalization, or even resulting in death. Gas can make an individual feel bloated and may cause abdominal pain, such as stomach cramps. Therefore, those experiencing this problem should make sure to replenish liquids whenever possible.
Examples of relaxing herb are German Chamomile and Marshmallow Leaf, which is both relaxing and soothing. Indeed, whereas non-pharmacological methods of obesity therapy demonstrate short-term efficacy (a loss of a number of kilograms in a couple of months), about half of that weight-loss is regained within one year and all the weight is regained within five years. Generic names are particularly useful in the world of research and education; one name, one molecule.
The mixture of bile salts and triacylglycerols leads to the formation of micelles in which the ester bonds of the triacylglycerols orient toward the surface. Low fat diets will cause less trouble (but the medicament becomes obsolete when you managed to eat less and eat less fat). Until today, bariatric surgery is considered only for morbid obese patients, who have not succeeded to reduce weight through life-style changes or medication, and for severe obese patients (MBI in between 35-40) with a high risk of morbidity and mortality. Advantages are: less post-operational pain, less scarring, minimal risk of incisional hernia, less trauma (faster recovery) and, perhaps, less (nosocomial) infection. Food slowly descents from the pouch, through a narrow exit, into the rest of the stomach, like in the hourglass depicted in figure 6a. The distal part of the duodenum, containing the ducts of the pancreas and the gallbladder (entry site of the digestive enzymes and the bile salts), is closed.
The secondary walls of woody tissue and grasses are composed predominantly of cellulose, lignin, and hemicellulose. In order to correct for the influence of pH on the fluorescent signal data was normalized using a fluorescence reference supplied by the assay kit at each pH.
The effect of temperature was measured by running separate assays at defined temperatures using the Synergy H4 Hybrid Multi- Mode Microplate Reader to maintain temperature. Subsequent data was normalized using the same factor and expressed as the change in signal of the initial unreacted fluorescence. Because microplates provide the ability to test large numbers of samples they are an ideal tool to test multiple experimental conditions. Towards that end, efforts have focused on ways to reduce the amount of acid used as a treatment by using a combination of dilute acids in conjunction with enzymatic hydrolysis. Because it is currently being used in large quantities the infrastructure to deliver the fuel already exists.
