Before you can do anything with DropSync you need to set up at least one Synced Place. Synced Places are like bookmarks for locations (folders, hard disks, networked computers etc) where you would like to store data. DropSync supports standard Synced Places (these are disks, or folders you can see in Finder) as well as ssh synced places (remote hosts accessible only through ssh).
