Creating a successful vaccine against two members of the family, the sexually transmitted herpes simplex virus 1 (HSV-1) and 2 (HSV-2), has proven to be challenging.
Meanwhile, Knipe and his colleagues are continuing to fight these viruses on another front.
Now Knipe and his colleagues have shown that compounds that block this crucial step in HIV infection also prevent genital herpes and other herpes virus infections from taking hold. The scientists tested the HIV integrase-inhibiting drug against herpes viruses, using it against HSV-1 and HSV-2 as well as the related cytomegalovirus and Kaposi's sarcoma herpes virus. An existing drug, acyclovir, effectively calms HSV-1 and HSV-2 outbreaks, but there are fewer choices for treating the reactivation of dormant cytomegaloviruses and Kaposi's sarcoma herpes viruses in people whose immune systems are compromised. Researchers have launched an early-stage clinical trial of an investigational vaccine designed to prevent genital herpes disease. The faster they come up with a vaccine that works, the better it will be for the general population.
Fluorescent staining shows areas of the cornea affected by herpes virus that infected a man’s eyes. Studies find that by age 60, virtually all adults carry herpes simplex virus 1 — best known for seeping cold sores but also potentially blinding when it hits the eyes.
It’s a tactic that researchers say may also hold promise for attacking HIV, another virus whose habit of hiding out makes it hard to kill, and the herpes zoster virus that causes excruciating shingles. Herpes viruses are best known for causing cold sores and genital lesions but they can also affect the eye, potentially leading to blindness. The antidepressant offers proof of concept, researchers say, but if the new method is eventually used on human patients, it’s likely to be in the form of new drugs developed specifically to act on LSD1.
Researchers are also trying to fight persistent viruses like HIV by trying to activate the virus; then they attack it with drugs or hope the immune system will clear it from the body.
But herpes viruses are reactivated in nerve cells, so they can potentially cause damage to the nervous system when they kick back up, says Harvard Medical School’s Dr. It uses a genetically engineered strain of the virus, he says, that can produce an infectious virus only in special cells in the laboratory.
The vaccine is meant to build immunity in people who have not yet caught the virus, but it may also work to suppress the virus somewhat in people who are already infected, Knipe says.
Kristie says he hopes the paper will stimulate HIV and other virus researchers to explore similar methods, and he sees potential spillover from cancer drugs that target epigenetics into other fields — like virus research. The report also indicates that  Duffy found only herpes simplex – 1 viruses (HSV-1) in the gastrointestinal tissues of the FM patients. After Pridgen noticed much greater improvement in the symptoms of the patients also put on Celebrex for their arthritis he combined the two drugs – for everyone.

That drug combo never been used in herpes virus infected patients before, but it made sense to Duffy. Valtrex, Valcyte and Vistide are often used to treat herpesvirus infections in chronic fatigue syndrome but Famvir is rarely mentioned. The group believes HSV-1 may be responsible for fibromyalgia, irritable bowel syndrome, chronic fatigue syndrome and perhaps other disorders. However, only 33% of patients (vs 19% of those not on it) met the criteria for a secondary measure called the  Global Impression of Change Scale which asked about changes in a person’s activity, limitation, symptoms, emotions and quality of life.
If the trial wasn’t a complete win it nevertheless produced significant improvements in a notoriously difficult to treat illness. Designed to  assess basic aspects of efficacy and safety, Phase II trials are not the last word in efficacy. Next up is the big third phase trial – a jaw dropping $50-100 million trial the FDA needs in order to approve the drug combo for the treatment of fibromyalgia. This is fascinating to me since I’ve had Herpes Simplex 1 infections in my eye (as well as fever blisters in the past) for fifty years now and had a major outbreak in my eye in the last year that coincided with more ME and Fibromyalgia symptoms. The very fact that these viruses are latent-residing, and are considered as parasites should have been a warning signal on the mainstream level ages ago.
I imagine that there may be so many longterm complications for some patients that even if HSV-1 is their triggering factor I imagine it may take more that removal of the virus to fully reverse the process.
Ive been following this story since the first trial and I have a bunch of questions which you may or may not have answers to. I suffer from fibromyalgia since 6 years and I consider asking my doctor (here in Germany) if he’d prescribe me these drugs for a self test. Hi Debbie, I finally found a doctor who would prescribe me these two drugs for off label use.
I don’t know how long a phase III trial will take but I imagine it will take quite some time. This is a very exciting, potential, treatment breakthrough, in my view, and I look forward to finding out the recommendations for dosages. My nervous system is being steadily degraded with autonomic nervous system problems, neuropathy and cognitive problems. A clinical trial being conducted by a branch of the National Institutes of Health, now enrolling patients, is based on an HSV-2 vaccine developed by David Knipe, Higgins Professor of Microbiology and Molecular Genetics at Harvard Medical School. Seizing an approach used against HIV, they are studying possible treatments that could also combat herpes virus infections.
They reported similarity between an HSV protein and the HIV integrase, which weaves a strand of viral DNA into a cell's chromosome, where it can then make copies of itself. Their results, published in July in the journal mBio, raise hopes for a new class of anti-herpes drugs that could potentially fend off both HIV and the herpes viruses. First it stops the virus from copying itself, and later it keeps it from producing the proteins needed to assemble new virus particles. When the virus destroys so many immune cells that the body can't fight off infection, AIDS will develop.

Herpes simplex virus 2, the sexually transmitted disease, infects more than a quarter of people by their forties, the CDC says. Their wily ways of going latent between recurrences, hiding out in viral reservoirs in our bodies, make them supremely hard to eradicate. Knipe was involved in developing a genital herpes vaccine that Harvard licensed to the pharmaceutical company Sanofi-Pasteur and that is now in initial safety trials at the National Institutes of Health. William “Skip” Pridgen and virologist Carol Duffy PhD used in their Fibromyalgia antiviral trial were Famciclovir, better known as Famvir and Celexicob, best known as Celebrex. Neither of these drugs nor this type of herpes virus have been commonly used or associated with chronic fatigue syndrome. Suspecting that herpes viruses might be to blame for the gastrointestinal issues in his patients, Pridgen started them off on Famvir.  The drug helped but symptoms remained. She knew that some herpes viruses  increase the production of COX-2, a pro- inflammatory enzyme.
A video on the Innovative Med Concepts website indicates the virus can attack the facial, gastrointestinal and pelvic regions and that it ultimately takes up residence in the vagus nerve. Why did these people get stuck with herpesvirus reactivation while others are able to fight it off. Also had that vagus nerve problem of vomiting in the morning due to the stomach not going to work on digestion when it should, but have remedied this by not eating until later in the day.
They screened similar compounds from an NIH library of HIV integrase inhibitors, coming up with some candidates that took much less drug to work better in herpes viruses. But Knipe says he has been thinking that it might first be tried in patients with recurrent herpes viruses that attack the eyes and can damage the cornea, leading to blindness. Your hosts are Carey Goldberg, former Boston bureau chief of The New York Times, and Rachel Zimmerman, former health and medicine reporter for The Wall Street Journal. While Famvir stopped the herpes viruses from replicating, Celebrex weakened the viruses, making them “unstable”. One article suggests herpes simplex virus may be better adapted to take advantage of poorly functioning natural killer cells than any other herpes virus.
